Autosomal dominant polycystic kidney disease (ADPKD), the most common form of polycystic kidney disease (PKD), is a genetic disorder in which fluid-filled cysts progressively develop in the kidney, leading to kidney damage and failure. Current treatments focus primarily on managing hypertension, the most common symptom of ADPKD. RAAS inhibitors, particularly lisinopril, dominate first-line therapy, and beta blockers, calcium channel blockers (CCBs), and diuretics compete for patient share in second-line therapy. Jynarque (tolvaptan) is the only disease-modifying therapy approved for ADPKD; it is used as first-line treatment in patients with rapidly progressing disease.
