Sjögren’s syndrome (SS) is an autoimmune disease characterized by dry eyes, dry mouth, and often also systemic manifestations such as fatigue and joint pain. While some SS cases are secondary, co-presenting with another, primary autoimmune disease, primary SS is the most common form. Current treatment of primary SS is mostly off-label and is focused on symptomatic relief of dry eyes, dry mouth, and/or systemic manifestations. Treatments that have a proven effect on autoimmune diseases other than SS have, for the large part, not demonstrated efficacy in SS, where the greatest unmet need remains for a single therapy that, likely by targeting an underlying disease process, could simultaneously address many of the syndrome’s signs and symptoms.
